{"created":"2025-03-12T08:11:14.527605+00:00","id":2000042,"links":{},"metadata":{"_buckets":{"deposit":"2911dafc-0bb5-4dab-9e29-5be113b3fbd9"},"_deposit":{"created_by":13,"id":"2000042","owner":"13","owners":[13],"pid":{"revision_id":0,"type":"depid","value":"2000042"},"status":"published"},"_oai":{"id":"oai:kyukan.repo.nii.ac.jp:02000042","sets":["1:4:1741746747621"]},"author_link":[],"control_number":"2000042","item_10002_biblio_info_7":{"attribute_name":"bibliographic_information","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2025-03","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"1","bibliographicPageEnd":"14","bibliographicPageStart":"2","bibliographicVolumeNumber":"25"}]},"item_10002_description_5":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"本研究では、糖尿病モデルラットから摘出した心臓を用いて、ノルエピネフリン付加低灌流障害および再灌流障害に対する外因性カルシトニン遺伝子関連ペプチド(Calcitonin gene-related peptide:CGRP)の作用を明らかにしようと試みた。血管の緊張は主に交感神経によって維持調節されている。一方、内因性CGRP は神経、特に知覚神経や血管周囲神経に存在し、強力な血管弛緩作用によって血管の緊張度を調節していることが示されている。\n 実験には、ストレプトゾトシン誘発6週間の糖尿病ラットおよび同週齢の非糖尿病ラットの摘出心臓を用い、ノルエピネフリン付加低灌流障害および再灌流障害に対する外因性CGRP の作用を比較検討した。心臓は電気刺激により300 beats/min で駆動し、5cm 水圧で通常灌流した後、30cm 水圧に下げ低流量灌流状態とし、再び75cm 水圧に戻し再灌流を行った。CGRP は低灌流時に9~10 nM、通常灌流時に2~3nM になるように調節し、低灌流にする15分前より実験終了まで持続注入した。低灌流5分後にノルエピネフリン(10-6 M)を含む灌流液に変え、低灌流45分後にノルエピネフリンを含まない灌流液で45分間の再灌流を行った。ノルエピネフリン付加によって、糖尿病心臓では低灌流障害、非糖尿病心臓では再灌流障害が亢進した。CGRP により、通常灌流時に糖尿病群・非糖尿病群の両群で、冠灌流量が有意に増加し、糖尿病群では、低灌流時に左心室収縮速度が上昇し、左心室スティフネスが増悪した。これらの結果より、ペーシング下における外因性CGRP の投与は、心臓の冠血管拡張作用を示したが、ノルエピネフリン付加低灌流障害および再灌流障害を増悪させることが示唆された。\nThis study aimed to determine the effects of exogenous calcitonin gene-related peptide (CGRP) on injuries induced by underperfusion with norepinephrine (NE) and reperfusion in hearts isolated from normal and streptozotocin-induced diabetic rats. Endogenous CGRP exhibits potent vasorelaxant effects and participates in the regulation of cardiovascular function. Although various factors are implicated in the cardiovascular dysfunction associated with the development of diabetes, the details of the mechanism remain poorly understood.Furthermore, studies with diabetic hearts have shown that diabetic hearts are more sensitive or resistant to ischemia-reperfusion injuries, which suggests that these discrepancies may be due to differences in the duration and degree of ischemia and the parameters monitored.\n For the experiments, the hearts of the streptozotocin-induced 6-week-old diabetic rats or age-matched nondiabetic rats were perfused and electrically paced in a Langendorff apparatus in the presence or absence of NE(10-6 M) and/or CGRP. The hearts were then subjected to 45 min of low-flow perfusion and 45 min of reperfusion.
